Abstract
Study Title: Oxygen Radical Absorbance Capacity - ORAC Activity of ION-ZCM1
Author: INDEVION Biotechnology Research and Development Ltd. Principal Investigator Dr. Zsolt Keresztessy, Ph.D. (2017).
Methods: In this investigative study for the antioxidant capacity of the test compound, ION-ZCM1. Using the antioxidant capacity of oxygen radicals (ORAC) assay system (OxiSelectTM kit an assay of the antioxidant capacity of oxygen radicals (ORAC) activity, Cell Biolabs). We measure the ORAC value of the ION-ZCM1. By definition, TOTAL ORAC =H-ORAC + L-ORAC, where H-ORAC means the ORAC capacity of the hydrophilic (water-soluble) fraction and L-ORAC means the ORAC capacity of the lipophilic (lipid-soluble) fraction. Since ION-ZCM1 is a water-based compound, it does not contain a lipid-soluble fraction. Therefore, when we determine the H-ORAC value of ION-ZCM1, we measure its TOTAL ORAC value, since the L-ORAC for ION-ZCM1 is zero (0), so the TOTAL ORAC = H-ORAC in the case of this compound.
Results: The results show that the H-ORAC of the ION-ZCM1 = TOTAL ORAC value (based on the standard TROLOX curve) is 740 μM of Trolox equivalents (TE) when measured at a 6.25% dilution, and is 1025 μM of Trolox equivalents (TE) when measured at a 12.5% dilution.
Conclusions: The ION-ZCM1 has a high ORAC value in comparison with other known substances, adding its already known redox potential of 450 mV and its direct absorption to the cell, which increases its total antioxidant activity. These factors place it at a very high level of value as therapeutic use. The comparison of absorption factors in food digestion against topical administration or other absorption methods adds to the complexity of accurate comparison. Further study of antioxidant activity in vivo provides a deeper understanding of possible therapeutic uses.
